it does not matter which PCI slot you use , however , make sure you install the drivers properly .
Most import, drivers should be compatible with your OS.

It *shouldn't* matter, but it might be a resource conflict problem. Is the card not detected at all, even in Device Manager?
I have found that it is usually best to NOT put the card in first.
If you have already installed the drivers - right click on the device in Device Manager and uninstall.
Shut down and take the card out.
Start back up and insert the CD. Cancel out of the auto setup and browse for the drivers ONLY. (I am assuming XP)
Install the drivers.
Shut back down.
Install the card.
Start back up and let Windows find the drivers and install.
It doesn't matter if a PCI port is working

It doesnt matter which slot it goes in as long as its configuerd connectly. USB is an esyerer way though I got a flash usb tuner for £25 uk british pounds.

No not relevent where it goes mate could be not compatible with your motherboard though don't install drivers until you plug it in make sure it's in all the way can sometimes pop back out. Then install the drivers.
