Completely depends on the video card itself. My card, a Radeon X1300 is PCI-E but has no use for any additional power other than provided by the PCI-E slot. Some high powered cards, like the 8800GTS, require additional power to be plugged in the the card. If you are considering pruchasing a high powered video card, usually more than $300, you better at least get a 500 Watt power supply. Most power supplies over 500 Watts will come with everything you need to run a single card. If you are planning on running dual video, or SLI, your cards AND power supply must support SLI function. the video card is powered by the mother board pcie slot and if the video card has a cooling fan on it you will need to have a power plug for it
Lower end cards in the class of a 7600GT and x1650 don't require a dedicated power connection (they draw from the PCI-e slot).
But the 7900gs and x1900 series and higher require the dedicated power connection in addition to the fan connector.
