yes if you format it the data will be gone..
most likely explanation is that your disk has experienced data corruption and its file allocation table is no longer readable...
run chkdsk [floppy drive]: /r and see if that works.. if not then I'm afraid your data is probably irretrievable.
Are you trying to Open it by going to MyComputer and clicking on the A:\ drive ??
Are you sure you have the right floppy disk ?
Are you sure when you placed the file on the disk it actually went to the disk ?
If you format it, yes you will lose the data .
It might be your floppy drive that is having a problem. See if anything is stuck inside the drive. Do not format it. Take it to another computer that has a working floppy drive and see if you can open it.

What system was used to store the file in the first place? The original format may be different from yours. Or, it may be that the disk was placed in closed proximity to a magnetic field and the information was corrupted or erased.
try clean your floppy drive lens first..
