Will a SATA II HHD, work on my motherboard's SATA port?

Yes, sata II will work on regular sata it will just run at the slower data rate which is around 150.
If your motherboard only supports the 1.5 GB SATA, you'll want to check the jumpers on the drive. I know OEM Seagate Barracudas come with the jumper set to the lower transfer rate and you need to pull the block to get the faster transfer rate. First off, check what the MoBo will handle (on the manufacturer's web site) and go from there. The connections and cables are the same for SATA or SATA II. I am a Seagate Partner Program Member.
So now check the HDD's manual to get the jumpers set correctly, if applicable. All the SATA drives I've worked with had jumpers, but perhaps not all do. Your mobo should automatically deal with whichever speed the HDD delivers.
you gave so little info... mention the mobo model!
as mentioned, check the mobo manual if you dont know what sata speed it handles, then check the hard drive manual to see if it will run at sata1 speeds (if the mobo is only capable for that)
